Gardaí said they and other emergency services were called to the crash scene around 1pm on the road between Buttevant and Liscarroll.
The driver of the van, a man in his late 30s, was pronounced dead at the scene a short time later. Two passengers in the van, described by gardaí as a male and female both in their late teens, were airlifted to Cork University Hospital. They are both reported to be in a serious condition.
The driver of the tractor, a man in his late 20s, was not physically harmed.

Forensic collision investigators were examining the crash site on Monday afternoon and the body of the van driver was taken to Cork University Hospital for a post-mortem investigation.
Gardaí are appealing for witnesses and anyone with information is asked to contact Mallow garda station on 022-31450 or the garda confidential line on 1800 666 111.
